Specimen Requirements
15 mL (minimum volume: 2 mL) from a first-morning urine collection is preferable, but testing can be performed on a random urine collection. Forward promptly. Specimen stability is 1 hour at ambient temperature. Specimens should be sent refrigerated if testing cannot occur within 1 hour of collection. Refigerated stability is 24 hours. Specimen should have no additive.

Clarity: clear
Glucose: negative mg/dL
Bilirubin: negative g/dL
Ketones: negative mg/dL
Specific gravity: 1.005-1.030
Blood: negative mg/dL
pH: 5.0-8.0
Protein: negative mg/dL
Urobilinogen: 0.1-1.0 eu/dL
Nitrites: negative mg/dL
Leukocyte esterase: negative
WBC: 0-5/HPF
RBC: 0-3/HPF
Squamous epithelial cells: 0-5/LPF
Note: A positive result is reported as a semi-quantitative number.
Reflex Notes
If ANY of the following criteria are met, a urine microscopic would be performed:
- Blood is positive (trace or greater)
- Protein is ≥30 mg/dL
- Nitrite is positive
- Leukoesterase is positive (trace or greater)
Note: Specimens collected in a urine bag or non-sterile container will not reflex to culture.
If the following criteria are met, a urine culture would be set-up:
- Microscopic WBC count >10/hpf
